Cancer cell lines, also known as tumor cell lines, are widely used as experimental models in cancer research. They are essential and reproducible model systems for understanding the development and characteristics of cancer, promoting the discovery and development of effective anticancer drugs. Due to their ease of manipulation and assessment, cancer cell lines are extensively used in both 2-dimensional (2D) and 3-dimensional (3D) cell culture systems. 2D culture brings more robust molecular profiling as the association with stromal and infiltrated immune cells are removed; while 3D culture gives the potential to mimic in vivo pharmacokinetics for drug testing. With technique advancements, such as omics technologies, a more comprehensive database is established based on cancer cell lines, which may help researchers to choose the appropriate models for their research.
