Ascent Research support various immortalized cells including spontaneous immortalized cell lines, and immortalized primary cells. Immortalized cells, or immortalized cell lines, are cells escaping replicate senescence through the spontaneous accumulation or artificial introduction of genetic/epigenetic mutations to inactivate growth arrest pathways, or activate telomerase (TERT), or bypass senescence and apoptosis checkpoints. Spontaneous immortalization requires continuous long-term culture, typically achieved through a process called serial transfer to select immortal clones. Other immortalization methods involve with transfection or transduction. Viral oncogenes (e.g. SV40 large T antigen, HPV E6/E7, Adenovirus E1A/E1B) are introduced to inactivate or degrade p53 and Rb, allowing bypass of senescence; or TERT is overexpressed to prevents telomere shortening, allowing cells to divide indefinitely.
