Immortalized cells or immortalized cell lines, are passaged or genetically modified primary cells to proliferate indefinitely in vitro. Immortalized cells overcome replicate senescence and obtain an extended lifespan through preventing telomeres shorten or inactivation of p53 and Rb. Spontaneously Immortalized Cell Lines

Spontaneously immortalized cell lines are cells that spontaneously acquire the ability to proliferate indefinitely without genetic manipulation or introduction of viral oncogenes. The spontaneous immortalization process rely on a long-term cell culture to accumulate mutations or epigenetic changes for bypassing replicate senescence and crisis stages.

Oncogene-immortalized Cell Lines

Oncogene-immortalized cell lines are immortal cell lines generated by the expression of viral oncogenes. These oncogenes are able to degrade or inactivate tumor suppressors, such as p53 and Rb, thereby bypassing senescence and apoptosis, or promote proliferation and metabolic reprogramming, allowing cells to proliferate indefinitely in vitro.

hTERT-immortalized Cell Lines

hTERT-immortalized cell lines are generated by introducing human telomerase reverse transcriptase (hTERT) gene into primary cells. This prevents telomere shortening to extended lifespan and retain a normal phenotype.

Hybrid Cell Lines

Hybrid cell lines are created by the fusion of two distinct cells. A common example is hybridoma, formed by fusing spleen cells (B cells) and myeloma cells for monoclonal antibody production.
