MAPDA knockout HT29 polyclonal cells are a CRISPR/Cas9-edited loss-of-function model targeting the scaffold protein MAPDA in human colorectal adenocarcinoma cells. Disruption of MAPDA, which facilitates MAPK signal transduction through interactions with MEK1 and ERK2, alters phosphorylation of downstream effectors such as ELK1 and c-Jun, impairing proliferation and survival signaling. These polyclonal cells derive from the HT29 intestinal epithelial line, widely used for colorectal cancer and drug transport studies, and are optimized for applications including Western blotting, apoptosis assays, and targeted therapy screening in MAPK-driven disease research.

Description

The MAPDA Knockout HT29 Polyclonal Cells product comprises a CRISPR/Cas9-edited polyclonal knockout population of the human colorectal adenocarcinoma cell line HT29, engineered to disrupt expression of the MAPDA gene.

This loss-of-function model provides researchers with a robust tool to interrogate the scaffold protein??s role in the mitogen-activated protein kinase (MAPK) signaling cascade.

By eliminating functional MAPDA, the cells enable dissection of kinase pathway architecture without the confounding presence of wild-type protein across a heterogeneous cell pool, reflecting the polyclonal nature of the knockout and avoiding artifacts associated with single-cell cloning.
