Description

Rigorous downstream applications are enabled by this knockout model, including quantitative Western blotting for ERBB2, phospho-AKT (Ser473), and phospho-ERK1/2 (Thr202/Tyr204) to validate pathway disruption.

Cell proliferation can be assessed via MTT or CCK-8 assays, while apoptosis is measured using annexin V/PI flow cytometry.

Transcriptional changes in ERBB2, MYC, and Cyclin D1 can be monitored by RT-qPCR, and ERBB2 subcellular localization can be examined via immunofluorescence.

The cells are also compatible with drug sensitivity screens for HER2-targeted therapeutics and with co-culture systems designed to explore B-cell immune functions.
