The KPNA1 Knockout Jurkat Polyclonal Cells are a CRISPR/Cas9?edited polyclonal population with disruption of the KPNA1 gene in Jurkat T lymphocytes. KPNA1 (importin ??5) is a key nuclear import adapter that recognizes classical NLS motifs on cargoes including NF???B p65, STAT1, p53, and viral proteins. Description

The KPNA1 Knockout Jurkat Polyclonal Cells are a CRISPR/Cas9-edited polyclonal population with disruption of the KPNA1 gene in Jurkat T lymphocytes. This polyclonal format provides a heterogeneous loss-of-function model, avoiding clonal biases while enabling effective perturbation of the classical nuclear import pathway. The cells are suitable for pooled functional genomics, high-content screening, and batch-to-batch consistency in biochemical assays.

The Jurkat cell line is a widely used human T lymphocyte model derived from acute T cell leukemia. It faithfully recapitulates T cell receptor signaling, apoptosis regulation, and cytokine responses, making it ideal for studying immune signaling networks and leukemogenesis. The line’s well-characterized signaling cascades facilitate a broad range of molecular and cellular analyses.

KPNA1 (importin ??5) is the nuclear import adapter for classical NLS-bearing cargoes. It binds NF-??B p65, STAT1, p53, and viral proteins and partners with importin ?? (KPNB1) to dock at the nuclear pore via nucleoporins NUP50 and NUP153. Import is driven by the Ran GTPase cycle and NTF2-mediated recycling. KPNA1 activity is regulated by cell cycle signals and stress-activated kinases, which may modulate cargo binding and translocation rates.

In Jurkat cells, KPNA1 disruption hinders nuclear import of transcription factors and viral proteins, impairing their downstream transcriptional programs. This model is valuable for probing the role of nuclear transport in T cell activation, survival, and transformation. Additionally, because KPNA1 is exploited by HIV-1 for nuclear entry of the pre-integration complex, these knockout cells offer a tool to dissect viral replication cycles and evaluate host-targeted antiviral strategies.

Applications include immunofluorescence tracking of mislocalized cargoes, co-immunoprecipitation of trafficking complexes, and reporter assays for NF-??B or STAT transcriptional activity. Flow cytometry can quantify apoptosis or cell cycle shifts. The cells are also suitable for screening classical import inhibitors and investigating viral-host interactions.
