CRISPR/Cas9-edited polyclonal NANS knockout Raji cells, a Burkitt??s lymphoma-derived B lymphocyte model, enable dissection of sialic acid biosynthesis and glycoimmunology. NANS disruption blocks conversion of N-acetylmannosamine 6-phosphate to N-acetylneuraminate 9-phosphate, impairing sialylation of glycoconjugates downstream of GNE and upstream of sialyltransferases such as ST6GAL1. Ideal for studying congenital disorders of glycosylation, B cell biology, and cancer glycosylation through lectin-based flow cytometry, sialic acid quantification, and cell adhesion assays.

NANS Knockout Raji Polyclonal Cells represent a CRISPR/Cas9-edited polyclonal cell population in which the NANS gene, encoding N-acetylneuraminic acid synthase, has been disrupted.

This knockout model provides a powerful loss-of-function tool for investigating sialic acid biosynthesis and its downstream effects on glycosylation-dependent processes.

The polyclonal format ensures a broad representation of editing outcomes across the population, avoiding single-clone bias and enabling robust phenotypic screening in a physiologically relevant immune cell background.
